The cytotoxic effects of methanol on the colony-forming capacity of six human hepatoma cell lines, Hep G2, Hep 3B, HLE, HuH-6, HuH-7 and PLC/PRF/5, were investigated. A closed system was used for the cytotoxicity assay, to prevent the rapid evaporation of methanol. The lethal dose of methanol for 50% cell death lay between 1% and 1.5%. Among the six hepatoma cell lines, Hep 3B and HuH-6 lines were a little more sensitive to methanol cytotoxicity than the other cell lines. Interestingly, alcohol dehydrogenase activity was inducible only in HuH-6 line after 24 hour exposure to 0.5% ethanol, though its activity was about one-half of that of the rat liver tissue.
